My sister and i would like to join the premium duo but we don't live together. Could this option be opened up to us?

Spotify Premium Family / Duo are offeres made for users who are either a family living under the same address or for 2 people living under the same address (Duo).

Unfortunately, sigining up requires an address confirmation which if does not match, the service will be unavailable.
