Sounds like you moved those local files on your PC. You can just add the folder in the desktop client preferences under Local Files/add source. Hopefully this will fix the greyed out tracks - you may need to restart spotify to force a refresh.
EDIT:You will probably need to resynch your playlists after doing that I'm afraid.

The files sit on an external hard drive and I think what happened was that it was disconnected at some point and in the shuffle got assigned a different letter.
That was so much easier of a solution than i was expecting.
