No that is not possible. iTunes gift cards can only be used for iTunes.

It should work the same way as Netflix does it through the iTunes app store:

In the Spotify iOS app you can pay for your subscription using your iTunes account and you might be able to use the gift card linked to your iTunes account. You will end up paying $12.99 per month instead of the $9.99 a month you can get from subscribing from the Spotify website (These are US prices, but international pricing is similar)
