I am using the Spotify app on windows 10. Will it not count as a stream if I lower the volume from the "app volume and device preferences" in sound settings of the laptop and keep the volume up on the app?

Absolutely! The only factor that's taken into account is how long you've been listening to the song, regardless of the playback volume. In general, it takes around 40-45 seconds of listening to count as one stream so feel free to listen as loud (or as quiet) as you like.
