how to query user profile for specific songs

Is it possible to see how often a user has listened to a specific song in their Spotify history using the web api?

Short answer: no.

Long answer: You can only get the last 50 played songs with the Get Recently Played Tracks endpoint. You can, however, get the current user's top tracks based on calculated affinity with the Get User's Top Items endpoint, but without amounts.
